The Mouse A673 Model is the most authoritative and widely adopted human-derived Ewing sarcoma (EWS) xenograft model for preclinical sarcoma research worldwide. A673 cells are derived from human primary Ewing sarcoma, carrying the typical EWS-FLI1 fusion gene and presenting high proliferation, strong invasion, poor differentiation and high malignant characteristics, which faithfully recapitulate the biological features of clinical bone and soft tissue Ewing sarcoma prevalent in children and adolescents. Logarithmic-phase A673 human sarcoma cells are subcutaneously inoculated into immunodeficient BALB/c-nu nude mice to establish stable and reproducible solid xenograft tumors without xenogeneic immune rejection. This model retains the core molecular phenotypes and malignant characteristics of human Ewing sarcoma, and is widely applied in exploring sarcoma proliferation, apoptosis, invasion, migration and targeted molecular mechanisms, as well as evaluating in vivo anti-tumor efficacy of small-molecule compounds, chemotherapeutic drugs and nano-formulations. Due to T-cell immunodeficiency in nude mice, this model is not suitable for immune mechanism research of complete immune checkpoint inhibitors, but only for intrinsic tumor biological research and preclinical drug screening. With mature system and stable tumorigenicity, it can be directly used for NSFC project application, postgraduate research proposals and standardized SCI manuscript methodology writing.
1. Positive modeling criteria: Subcutaneous tumor nodules are visible to the naked eye within 7–12 days after A673 cell subcutaneous inoculation. The tumor formation rate is ≥90% within 2 weeks after inoculation, and local tumor volume >50 mm³ is defined as successful modeling. Tumors in the vehicle control group grow continuously and stably without spontaneous regression or atrophy, which conforms to the continuous proliferation characteristics of Ewing sarcoma in vivo. 2. Model reproducibility criteria: The intra-group coefficient of variation (CV) of tumor volume in the vehicle control group is <30%, which proves standardized modeling operation, small individual differences and excellent model data reproducibility, meeting the quality control requirements of SCI experimental data. 3. Pharmacodynamic response criteria: After intervention with positive control drugs, the tumor growth inhibition rate (TGI) is ≥40%, and tumor volume and tumor weight are significantly reduced compared with the vehicle control group (P<0.05), verifying that the model is sensitive to chemotherapy and targeted drug intervention with a stable and reliable pharmacodynamic evaluation system.
1. HE staining gold standard: A large number of densely arranged small round blue cells with large nuclei, little cytoplasm, significant nuclear atypia and active mitotic figures are observed in tumor tissues, which are completely consistent with the typical pathological morphological characteristics of human Ewing sarcoma. 2. Immunohistochemical phenotypic criteria: High expression of Ki-67 and PCNA in tumor tissues indicates high tumor proliferative activity, and positive expression of MMP2 and MMP9 suggests a high-invasive sarcoma phenotype, perfectly recapitulating the core biological characteristics of malignant proliferation and high invasion of clinical Ewing sarcoma. 3. Pharmacodynamic response criteria: Compared with the vehicle control group, the treatment group shows significantly down-regulated proliferation markers (Ki-67, PCNA), significantly up-regulated apoptosis marker (Cleaved-Caspase-3), and obviously decreased invasion markers (MMP2, MMP9), indicating that the drug can effectively inhibit the proliferation and invasion of Ewing sarcoma and activate tumor cell apoptosis. 4. Evidence closed-loop criteria: In vivo histopathological and molecular protein detection results are highly consistent with the in vitro A673 cell experimental mechanism trend, forming a complete and credible academic evidence closed loop.
The Mouse A673 Model is a recognized classic gold-standard in vivo model for preclinical Ewing sarcoma research. The core academic validity evidence: ① The cell line has specific fusion gene phenotype of human Ewing sarcoma with typical malignant characteristics, which is consistent with clinical disease features; ② The nude mouse xenograft system has stable tumorigenesis, high success rate and good data reproducibility; ③ It can stably respond to chemotherapy, targeted drugs and small-molecule drug intervention with clear pharmacodynamic gradients; ④ The indicator system covers multiple dimensions including proliferation, apoptosis, invasion, pathology, toxicology and molecular pathways with a complete evidence chain; ⑤ Experimental data are widely recognized by global SCI journals in the fields of sarcoma and orthopedic oncology. It is a core model for Ewing sarcoma mechanism research and preclinical transformation of new drugs, suitable for various project applications, research proposals and SCI manuscript methodology writing.
The Mouse A673 Model is the most classic and widely used preclinical in vivo model for human Ewing sarcoma with prominent core advantages. The cell line retains the characteristic fusion gene and small round cell malignant phenotype of Ewing sarcoma, which is highly consistent with the pathological and biological characteristics of clinical Ewing sarcoma. Human-derived tumor xenograft avoids species differences and is closer to human diseases compared with mouse syngeneic sarcoma models. Subcutaneous modeling has the advantages of simple operation, short cycle, extremely stable tumorigenesis and small individual differences. It is sensitive to clinical first-line anti-sarcoma drugs with clear pharmacodynamic gradients, suitable for high-throughput drug screening and mechanism verification. The comprehensive indicator system can simultaneously evaluate tumor proliferation, apoptosis, invasion and in vivo drug toxicity. With a mature system and highly recognized data, it is fully compatible with NSFC projects, provincial and ministerial projects, postgraduate research proposals, dissertations and SCI methodology writing, and can form a complete tumor research system with other solid tumor models of the research group.
The Mouse A673 Model is mainly used forbasic mechanism research and preclinical translational research of human Ewing sarcoma. Core application scenarios include: mechanism analysis of downstream proliferation, invasion and apoptosis signaling pathways regulated by EWS-FLI1 fusion gene in Ewing sarcoma; in vivo anti-Ewing sarcoma efficacy evaluation of chemotherapeutic drugs, targeted small-molecule drugs, natural products and nano-formulations; research on the regulatory mechanism of malignant biological behavior of sarcoma; verification of the in vivo efficacy enhancement and toxicity reduction effects of new combined therapeutic regimens; preclinical new drug screening and mechanism verification for orthopedic malignant tumors. It fully supports the methodology construction of NSFC projects, research proposals, dissertations and SCI papers in the field of urinary system and bone and soft tissue tumors, serving as an irreplaceable SCI gold-standard in vivo model in the field of Ewing sarcoma research.
