Modeling Principle

The Mouse C666-1 Model is a exclusive gold-standard in vivo model for preclinical research on **EBV-positive, highly invasive and metastatic human nasopharyngeal squamous cell carcinoma**. Derived from human primary nasopharyngeal carcinoma, C666-1 cells are one of the few stably EBV-infected human NPC cell lines, faithfully retaining the core malignant phenotypes of clinical NPC including EBV positivity, high invasion, high metastasis, easy recurrence and radioresistance. Compared with conventional CNE NPC cell lines, C666-1 presents stronger epithelial-mesenchymal transition (EMT) activity, in vitro migration and invasion capacity, and in vivo distant metastatic potential, which highly conform to the tumor biological characteristics of clinically prevalent, highly metastatic and poor-prognosis EBV-positive nasopharyngeal carcinoma. Logarithmic-phase C666-1 human NPC cells are subcutaneously inoculated into BALB/c-nu immunodeficient nude mice to establish a stable subcutaneous xenograft model with prominent metastatic phenotypes and negligible batch variation without xenogeneic immune rejection. This model is widely applied in the research of NPC malignant proliferation, EMT-mediated invasion and metastasis, EBV-driven oncogenic pathways, radioresistance and cancer stemness regulation. It is also suitable for in vivo efficacy evaluation of small-molecule targeted drugs, natural bioactive compounds, nano-delivery systems and sensitizing agents. With a mature system and high SCI recognition, it fully meets the methodological writing requirements of NSFC projects, postgraduate proposals, dissertations and SCI manuscripts.

Model Validation Criteria

Quantitative Criteria for Tumor Formation and Growth

1. Positive modeling criteria: Clear macroscopic tumor nodules appear 6–10 days after C666-1 cell subcutaneous inoculation. The tumor formation rate is ≥92% within 12 days, and tumor volume >50 mm³ is defined as successful modeling. Tumors in the vehicle control group proliferate continuously and stably without spontaneous regression with good growth homogeneity, which are completely consistent with the high-proliferation and high-invasion malignant growth characteristics of clinical EBV-positive nasopharyngeal carcinoma.

2. Model reproducibility criteria: The intra-group coefficient of variation (CV) of tumor volume in the vehicle control group is <30%, indicating high standardized modeling operation, small individual differences and excellent batch stability, fully meeting the quality control requirements of SCI experimental data.

3. Pharmacodynamic response criteria: After intervention with NPC positive control drugs, the tumor growth inhibition rate (TGI) is ≥40%, and tumor volume and tumor weight are significantly reduced compared with the vehicle control group (P<0.05), proving that the model has stable and sensitive pharmacodynamic response to chemotherapy, targeted drugs and sensitizers, suitable for NPC drug screening and mechanism verification.

Histopathological and Molecular Gold-Standard Indexes

1. HE staining gold standard: Tumor tissues contain a large number of severely atypical nasopharyngeal squamous carcinoma cells with disordered arrangement, large deeply stained nuclei, abundant mitotic figures and high cellular density, with local invasive growth characteristics, which are completely consistent with the typical pathological manifestations of human highly metastatic nasopharyngeal squamous cell carcinoma.

2. Molecular phenotypic criteria: High Ki-67 expression indicates strong proliferative activity; high expression of N-cadherin, Vimentin, MMP2 and MMP9 combined with low E-cadherin expression confirms significant EMT activation and high invasion and metastasis phenotypes; high CD133 expression indicates tumor stemness and treatment resistance characteristics, perfectly recapitulating the core malignant molecular phenotypes of clinical EBV-positive nasopharyngeal carcinoma.

3. Pharmacodynamic response criteria: Compared with the vehicle control group, the treatment group shows significantly down-regulated Ki-67, significantly up-regulated Cleaved-Caspase-3, recovered E-cadherin, decreased N-cadherin/Vimentin/MMP2/MMP9 and weakened CD133 expression, confirming that drugs can effectively inhibit NPC proliferation, promote tumor apoptosis, reverse EMT-mediated invasion and metastasis, and attenuate tumor stemness.

4. Evidence closed-loop criteria: In vivo histopathological and molecular protein detection results are highly consistent with the in vitro C666-1 cell experimental mechanism trend, forming a complete and credible academic evidence closed-loop.

Academic Evidence for Model Validity

The Mouse C666-1 Model is the only exclusive gold-standard in vivo model for preclinical research on EBV-positive highly metastatic nasopharyngeal carcinoma. Core academic validity evidence: ① The cell line stably carries Epstein-Barr virus, accurately corresponding to the most common and worst-prognosis EBV-associated NPC subtype in clinic, which is different from conventional EBV-negative NPC models; ② It possesses malignant phenotypes of strong EMT activation, high invasion and metastasis, high stemness and radioresistance, highly simulating the core clinical problems of NPC recurrence, metastasis and treatment resistance; ③ Subcutaneous xenograft presents stable tumorigenesis without loss of malignant phenotypes and minimal batch variation, suitable for long-term research on metastasis mechanisms and sensitization strategies; ④ It is sensitive to chemotherapy, targeted drugs, natural products and radiotherapy sensitization intervention with clear pharmacodynamic gradients; ⑤ Experimental data are widely recognized by head and neck oncology SCI journals. As an irreplaceable core model for EBV oncogenic mechanism, NPC metastasis and stemness, radiotherapy sensitization and drug resistance reversal research, it is applicable for methodology construction of various projects, research proposals and high-score papers.

Model Advantages

The Mouse C666-1 Model is an exclusive SCI gold-standard model for mechanism and translational research of EBV-positive highly metastatic nasopharyngeal carcinoma. Core advantages: As the most clinically relevant in vivo model with stable EBV positivity in China, it perfectly simulates the real clinical characteristics of NPC including virus-mediated carcinogenesis, high invasion, high metastasis, easy recurrence and radioresistance. With strong EMT metastatic phenotype and stable in vivo malignant progression, it serves as the preferred model for NPC metastasis mechanism, stemness regulation and treatment sensitization research. It features high tumor homogeneity, small individual differences, moderate experimental cycle and excellent data reproducibility, suitable for in-depth exploration of innovative mechanisms for high-score papers and project innovation design. It can form a control system with conventional EBV-negative NPC models to realize differentiated research on virus-mediated carcinogenic mechanisms. It constructs a multi-tumor research matrix with the group’s bladder cancer models, improving project breadth and innovation with excellent recognition for NSFC projects and SCI manuscripts.

Research Applications

The Mouse C666-1 Model is mainly used for preclinical mechanism and translational research on malignant progression, EMT metastasis, tumor stemness and radioresistance of EBV-positive highly metastatic nasopharyngeal carcinoma. Core application scenarios: mechanism analysis of EBV-mediated activation of NPC oncogenic pathways; research on EMT invasion and metastasis, abnormal stemness activation and recurrent drug resistance mechanisms of nasopharyngeal carcinoma; in vivo anti-tumor, anti-metastasis and sensitization efficacy evaluation of small-molecule targeted drugs, natural bioactive products and nano-formulations against highly metastatic NPC; development of radiotherapy/chemotherapy sensitization strategies and exploration of drug resistance reversal mechanisms for NPC; research on differential pathogenic mechanisms and targeted therapy of virus-related tumors. It fully supports innovative mechanism exploration of NSFC projects, postgraduate proposals, dissertations and high-score SCI papers in head and neck oncology, serving as an indispensable core gold-standard in vivo model for basic and translational research of EBV-positive nasopharyngeal carcinoma.
