Squamous Cell Carcinoma of the Tonge

Modeling Method

Subcutaneous

Verifacition

Model Establishment Principle

CAL‑27 is a human tongue squamous cell carcinoma cell line with stable proliferative capacity and retains the core malignant biological phenotype of tongue squamous cell carcinoma. Subcutaneous xenograft tumors can be established in immunodeficient mice. This model avoids the species difference of syngeneic tumors, mimics the in vivo growth and invasion characteristics of human tongue squamous cell carcinoma, and is widely used for in vivo efficacy evaluation of oral squamous cell carcinoma pathogenesis, anti‑tumor candidate drugs, gene therapy and biomaterials.

Model Identification & Evaluation Indicators

Gross observation: The tumor long diameter (a) and short diameter (b) are measured with vernier calipers every 2–3 days after inoculation. Tumor volume formula: V = 0.5 × a × b². Record the changes of mouse body weight. When the tumor volume reaches 100–150 mm³, the model is considered successfully established, and grouping and drug intervention can be carried out.

Histopathological examination: Mice are sacrificed at the endpoint of the experiment, tumor tissues are isolated, fixed in 4% paraformaldehyde, embedded in paraffin and sectioned. HE staining is performed to observe tumor histomorphology; Immunohistochemistry is used to detect proliferation markers such as Ki‑67 and PCNA to verify the malignant phenotype of tumor.

Molecular biological detection: Tumor tissue samples are collected for qPCR and Western blot to detect the expression levels of tumor‑related genes and proteins.

Model Advantages

The CAL‑27 mouse xenograft model has stable tumor formation rate and good repeatability. The cells are human tongue squamous cell carcinoma cells, which are closer to the biological characteristics of human tongue cancer compared with syngeneic oral squamous cell carcinoma mouse tumors. Subcutaneous modeling is easy to operate, and the tumor is superficially located, which facilitates non‑invasive measurement and dynamic monitoring. It is suitable for in vivo anti‑tumor drug screening and mechanism research.

Typical Application Scenarios

It is applied to the study of in vivo proliferation and invasion mechanisms of human tongue squamous cell carcinoma; in vivo anti‑tumor activity evaluation of small‑molecule targeted drugs, chemotherapeutic drugs, nanoformulations and gene delivery systems; preclinical pharmacodynamic verification of oral tumor biomaterials and combined therapeutic strategies.
