Modeling Principle

The Mouse HT‑1080 Model is a globally recognized high‑reproducibility gold‑standard xenograft model for human soft‑tissue fibrosarcoma research. Derived from human malignant fibrosarcoma, HT‑1080 cells possess strong proliferative, tumorigenic, invasive and pulmonary metastatic capacities with stable malignant phenotypes, which faithfully recapitulate the progression and metastatic biological characteristics of clinical highly invasive human soft‑tissue fibrosarcoma. Logarithmic‑phase HT‑1080 human fibrosarcoma cells are subcutaneously inoculated into immunodeficient BALB/c‑nu nude mice to establish stable, homogeneous and batch‑repeatable solid xenograft tumors without xenogeneic immune rejection. This model retains the intrinsic high‑proliferation, high‑invasion and high‑metastasis phenotypes of human fibrosarcoma, and is widely applicable for exploring soft‑tissue sarcoma proliferation, apoptosis, epithelial‑mesenchymal transition, matrix remodeling, invasion and metastasis mechanisms, as well as evaluating in‑vivo anti‑sarcoma efficacy and safety of small‑molecule targeted drugs, chemotherapeutic agents, natural products and nano‑delivery systems. Due to the absence of functional T‑lymphocytes in nude mice, this model is not suitable for adaptive immune checkpoint inhibitor studies, but only for intrinsic tumor biological research and non‑immune‑dependent drug evaluation. With a mature system and highly credible data, it can be directly applied to NSFC project applications, postgraduate proposals, dissertations and standardized SCI methodology writing.

Model Validation Criteria

Quantitative Criteria for Tumor Formation and Growth

1. Positive modeling criteria: Subcutaneous tumor nodules are visible 5–10 days after HT‑1080 cell inoculation. The tumor take rate is ≥92% within 2 weeks, and tumor volume >50 mm³ is defined as successful modeling. Tumors in the vehicle control group grow continuously and stably without spontaneous regression, consistent with the high‑proliferation characteristics of human fibrosarcoma in vivo. 2. Reproducibility criteria: The intra‑group coefficient of variation (CV) of tumor volume in the vehicle group is <30%, indicating good modeling uniformity and low systematic error, which meets SCI quality control requirements for data repeatability. 3. Pharmacodynamic response criteria: Positive drug intervention achieves TGI≥40% with significantly reduced tumor volume and weight compared with the control group (P<0.05). The model presents stable and sensitive responses to chemotherapy and targeted drugs with a reliable pharmacodynamic evaluation system.

Histopathological and Molecular Gold-Standard Indexes

1. HE gold standard: Tumor tissues are composed of massive disorderly arranged spindle‑shaped fibrosarcoma cells with large deeply stained nuclei, significant nuclear atypia, abundant mitotic figures and obvious stromal hyperplasia, which are completely consistent with the typical pathological features of human fibrosarcoma. 2. Molecular phenotypic criteria: High Ki‑67 expression indicates strong proliferative activity; high Vimentin and low E‑cadherin expression indicate significant EMT mesenchymal transition phenotype; high MMP2/MMP9 expression verifies strong invasion and metastasis potential, perfectly recapitulating the malignant phenotype of clinical soft‑tissue fibrosarcoma. 3. Pharmacodynamic response criteria: Treatment groups show down‑regulated Ki‑67, up‑regulated Cleaved‑Caspase‑3, decreased MMP2/MMP9, reduced Vimentin and recovered E‑cadherin, confirming that drugs can significantly inhibit proliferation, promote apoptosis, reverse EMT and weaken invasion and metastasis capacity. 4. Evidence closed‑loop criteria: In vivo tissue detection results are highly consistent with in vitro HT‑1080 cell mechanistic experiments, forming a complete and credible academic evidence chain.

Academic Evidence for Model Validity

The Mouse HT‑1080 Model is a recognized gold‑standard preclinical model for human soft‑tissue fibrosarcoma research. Core validity evidence: ① The cell line belongs to human malignant fibrosarcoma lineage with high malignancy and typical invasion‑metastasis characteristics, which is highly consistent with clinical soft‑tissue sarcoma features; ② Nude mouse xenograft presents extremely high tumor take rate, strong stability and minor batch variation, suitable for high‑throughput pharmacodynamic screening; ③ It possesses multi‑dimensional phenotypes including proliferation, invasion, EMT, matrix remodeling and metastasis with comprehensive mechanistic research dimensions; ④ It responds sensitively and gradiently to chemotherapy, targeted drugs and natural product intervention; ⑤ Its data are widely accepted by global SCI journals in sarcoma, orthopedics and oncology, serving as a core in‑vivo model for fibrosarcoma mechanism exploration and new drug transformation, fully applicable for project applications, research proposals and manuscript methodology writing.

Model Advantages

The Mouse HT‑1080 Model is the most classic, stable and widely applied in‑vivo model for human fibrosarcoma research. Core advantages: Human‑derived tumor cells completely avoid species deviation of mouse syngeneic sarcoma and highly recapitulate the malignant proliferation, stromal invasion, EMT transformation and distant metastasis characteristics of clinical soft‑tissue fibrosarcoma. It features fast tumor formation, extremely high tumor take rate, minor individual differences and short experimental cycle. With abundant phenotypes, it supports multi‑dimensional studies on proliferation, apoptosis, stromal invasion, EMT and metastasis mechanisms. It is sensitive to chemotherapy, targeted drugs and natural small‑molecule compounds with clear pharmacodynamic gradients and stable data. The mature system is globally recognized, and its methodology can be directly applied to SCI manuscripts and project applications. It maintains fully unified formats, parameters and quality control with 786‑O, A673 and CAL‑27 models of the research group, facilitating horizontal multi‑model comparative studies.

Research Applications

The Mouse HT‑1080 Model is mainly applied to basic mechanism research and preclinical translational research of human soft‑tissue fibrosarcoma. Core application scenarios: exploration of molecular mechanisms underlying fibrosarcoma proliferation, apoptosis, matrix remodeling, EMT and invasion‑metastasis; in‑vivo anti‑sarcoma efficacy evaluation of chemotherapeutic drugs, targeted small molecules, natural products and nano‑carrier formulations; verification of efficacy enhancement and toxicity reduction of combined therapeutic strategies for soft‑tissue sarcoma; research on sarcoma malignant progression and microenvironment interaction mechanisms. It supports methodology construction for NSFC and provincial‑level projects, postgraduate proposals, dissertations and SCI papers in the field of orthopedic and soft‑tissue tumors, serving as an indispensable standard SCI in‑vivo model in soft‑tissue sarcoma research.
