Modeling Principle

The Mouse MB49 Model is the most authoritative and highly adaptable syngeneic orthotopic bladder cancer gold-standard model in urinary system oncology. Derived from spontaneous bladder urothelial carcinoma of C57BL/6 mice, MB49 cells possess a fully homologous genetic background with C57BL/6 mice, stable proliferation, definite invasive capacity and typical malignant urothelial phenotypes, which faithfully recapitulate the in-situ growth, invasive progression and immune response characteristics of clinical primary bladder cancer. MB49 cells are instilled into the mouse bladder cavity via the urethra to establish an immune-competent orthotopic bladder cancer model. Different from immunodeficient xenograft models, this system retains intact host immune system and native bladder anatomical structure, truly reproducing bladder tumor microenvironment, tumor-immune interaction and in vivo responses to natural products, targeted drugs, chemotherapy and immune checkpoint inhibitors. With excellent data reproducibility and high SCI recognition, the MB49 model serves as a core platform for bladder cancer mechanism exploration, immunotherapy verification and preclinical drug screening, fully applicable to NSFC projects, postgraduate proposals, dissertations and standardized SCI methodology writing.

Model Validation Criteria

Quantitative Criteria for Tumor Formation and Growth

1. Positive modeling criteria: Stable bladder tumor colonization occurs within 7 days after MB49 orthotopic instillation, with visible bladder wall thickening, local protuberance and mild gross hematuria. The tumor formation rate is ≥95% on day 14 after modeling without spontaneous regression, which is defined as successful modeling. 2. Reproducibility criteria: The intra-group coefficient of variation (CV) of tumor burden, hematuria degree and body weight change in the model group is <30%, indicating excellent modeling uniformity and low systematic error, meeting SCI data quality control requirements. 3. Pharmacodynamic response criteria: Positive drug intervention significantly reduces tumor burden, relieves hematuria symptoms and alleviates bladder invasion with statistically significant differences compared with the model group (P<0.05), verifying that the model has stable and sensitive responses to chemotherapy, targeted therapy and immunotherapy.

Histopathological and Molecular Gold-Standard Indexes

1. HE gold standard: Bladder tissues show urothelial dysplasia, disordered arrangement, large deeply stained nuclei and active mitotic figures. Tumor cells infiltrate the mucosal and submucosal layers with local inflammatory cell infiltration, which is completely consistent with the typical pathological features of mouse orthotopic urothelial carcinoma. 2. Molecular phenotypic criteria: High Ki-67 and PCNA expression indicates high proliferative activity; high MMP2/MMP9 expression suggests invasive phenotype of bladder tumors; normal infiltration of CD8+ T cells and macrophages in the model group confirms intact tumor immune microenvironment. 3. Pharmacodynamic response criteria: Treatment groups show down-regulated proliferation markers, up-regulated apoptosis markers, decreased invasion indicators and enhanced immune infiltration, verifying that drugs can inhibit bladder cancer proliferation and invasion, activate tumor apoptosis and reshape tumor immune microenvironment. 4. Evidence closed-loop criteria: In vivo pathological and molecular detection results are highly consistent with in vitro MB49 cell experimental mechanisms, forming a complete and credible academic evidence chain.

Academic Evidence for Model Validity

The Mouse MB49 Model is a recognized immune-competent orthotopic gold-standard model in urinary bladder cancer research. Core academic validity evidence: ① The cell line is genetically syngeneic with mice without xenogeneic rejection, and the microenvironment is more consistent with clinical orthotopic bladder cancer; ② It retains intact adaptive and innate immune systems, serving as the preferred model for bladder cancer immunotherapy and immune microenvironment mechanism research; ③ Orthotopic colonization completely simulates the pathogenesis and invasive progression of clinical bladder cancer; ④ It presents an extremely high tumor formation rate, strong stability and minor batch variation, suitable for high-throughput pharmacodynamic screening and mechanism verification; ⑤ It supports multi-dimensional evaluation of chemotherapy, targeted therapy, immunotherapy and combined treatment. Its data are widely accepted by global urological oncology SCI journals, fully applicable for project applications, research proposals and manuscript methodology writing.

Model Advantages

The Mouse MB49 Model is the most authoritative and widely used immune-competent orthotopic model for bladder cancer research. Core advantages: Syngeneic cells combined with wild-type C57BL/6 mice have no immune deficiency and xenogeneic rejection, perfectly retaining the tumor-immune interactive microenvironment, which makes it the only standardized in-vivo model suitable for bladder cancer immunotherapy mechanism research. Intravesical orthotopic colonization completely recapitulates the disease characteristics of clinical urothelial carcinoma including pathogenesis, local invasion and hematuria progression. It achieves nearly 100% tumor formation rate with stable modeling, minor individual differences and controllable experimental cycle, supporting multi-dimensional pharmacodynamic evaluation of chemotherapy, targeted therapy, immunotherapy and combined treatment. With a mature system and top global SCI recognition, it forms a complete tumor research matrix of "immune-deficient + immune-competent, ectopic + orthotopic" with previous xenograft models, suitable for innovative project design and in-depth mechanism exploration for high-score papers.

Research Applications

The Mouse MB49 Model is mainly applied to in-situ mechanism, tumor immunity and preclinical translational research of bladder urothelial carcinoma. Core application scenarios: exploration of in-situ proliferation, mucosal invasion and apoptosis imbalance mechanisms of bladder cancer; evaluation of bladder tumor immune microenvironment regulation, immune escape mechanisms and immune checkpoint inhibitor efficacy; in-vivo anti-bladder cancer efficacy and safety verification of single and combined regimens of chemotherapy, targeted drugs, natural products and nanoformulations; preclinical screening and mechanism exploration of novel therapeutic strategies for urinary system tumors. It supports methodology and innovation construction for NSFC and provincial-level projects, postgraduate proposals, dissertations and high-score SCI papers in urological oncology, serving as an indispensable immune gold-standard in-vivo model in bladder cancer research.
