Modeling Principle

The Mouse OE19 Model is a preclinical gold-standard in vivo model for **HER2-positive esophagogastric junction (EGJ) adenocarcinoma** in gastrointestinal oncology. Derived from human moderately differentiated adenocarcinoma of the esophagogastric cardia junction, OE19 (JROECL19) cells are a exclusive cell line with stable HER2 overexpression, consistent malignant phenotypes and extremely high in vivo tumorigenicity. It faithfully recapitulates the core pathological features of clinical stage III EGJ adenocarcinoma, including vigorous proliferation, strong invasiveness, frequent metastasis, HER2-targeted dependence and recurrent drug resistance. Compared with conventional esophageal cancer cell lines, OE19 presents definite HER2 gene amplification and protein overexpression, remarkable EMT activation, rapid in vivo proliferation and homogeneous malignant progression, which perfectly matches the research scenarios of clinical advanced HER2-positive EGJ adenocarcinoma treatment and drug resistance. Logarithmic-phase human OE19 esophageal adenocarcinoma cells are subcutaneously inoculated into BALB/c-nu immunodeficient nude mice to establish subcutaneous solid tumors with no immune rejection, stable tumorigenesis and high phenotypic fidelity. This model is dedicated to the research of malignant proliferation, invasion and metastasis, HER2-driven oncogenic pathways and targeted resistance mechanisms of EGJ adenocarcinoma. It is also applicable for in vivo efficacy evaluation of trastuzumab, small-molecule HER2 inhibitors, chemotherapy sensitizers, natural products and nano-formulations. With excellent data reproducibility and high SCI recognition, it fully meets the methodological writing requirements of NSFC projects, postgraduate proposals, dissertations and SCI manuscripts.

Model Validation Criteria

Quantitative Criteria for Tumor Formation and Growth

1. Positive modeling criteria: Clear subcutaneous tumor nodules appear 5–7 days after OE19 cell inoculation. The tumor take rate is ≥95% within 10 days, and tumor volume >50 mm³ is defined as successful modeling. Tumors in the vehicle control group grow rapidly and continuously without spontaneous regression, which is completely consistent with the high-proliferation and high-malignant progression characteristics of clinical HER2-positive esophagogastric junction adenocarcinoma.

2. Reproducibility criteria: The intra‑group coefficient of variation (CV) of tumor volume in the vehicle group is <30%, indicating good modeling uniformity and low systematic error, which strictly meets SCI quality control requirements for data repeatability.

3. Pharmacodynamic response criteria: HER2-targeted positive drug intervention achieves TGI≥45% with significantly reduced tumor volume and weight compared with the control group (P<0.05). The model presents stable and sensitive responses to targeted drugs and chemotherapeutic drugs, enabling accurate evaluation of targeted sensitization and drug resistance reversal effects in esophageal adenocarcinoma.

Histopathological and Molecular Gold-Standard Indexes

1. HE gold standard: Tumor tissues consist of a large number of atypical esophageal adenocarcinoma cells with disordered arrangement, abnormal glandular structure, large deeply stained nuclei, abundant mitotic figures and high cellular density, which conform to the typical pathological morphological characteristics of human moderately differentiated esophagogastric junction adenocarcinoma.

2. Molecular phenotypic criteria: **Stable high HER2 protein expression** in tumor tissues serves as the core gold standard of the model. High Ki‑67 expression indicates strong proliferative activity; high Vimentin, MMP2 and MMP9 expression combined with low E‑cadherin expression confirms significant EMT and high invasive phenotype; high CD44 expression indicates tumor stemness and targeted drug resistance potential, perfectly recapitulating the core molecular characteristics of clinical HER2-positive esophageal adenocarcinoma.

3. Pharmacodynamic response criteria: Treatment groups show down-regulated HER2 expression, decreased Ki‑67, up-regulated Cleaved‑Caspase‑3, weakened MMP2/MMP9, recovered E‑cadherin and reduced CD44 expression, confirming that drugs can effectively inhibit tumor proliferation, promote apoptosis, block invasion and metastasis, down-regulate HER2 pathway and reverse targeted drug resistance.

4. Evidence closed‑loop criteria: In vivo tissue detection results are highly consistent with in vitro OE19 cell mechanistic experiments, forming a complete and credible evidence chain of targeted intervention mechanisms.

Academic Evidence for Model Validity

The Mouse OE19 Model is an internationally recognized gold-standard in vivo model for targeted therapy and drug resistance research of HER2-positive esophagogastric junction adenocarcinoma. Core validity evidence: ① Derived from clinical esophagogastric junction adenocarcinoma, the cell line stably harbors HER2 amplification phenotype and serves as the most authoritative and widely used cell model for HER2-targeted research in esophageal adenocarcinoma; ② It presents an extremely high in vivo tumor formation rate and stable malignant phenotypes without phenotypic loss, accurately simulating the pathological progression of proliferation, invasion and primary/secondary targeted resistance in clinical advanced esophageal adenocarcinoma; ③ It has clear gradient pharmacodynamic responses to HER2 inhibitors, chemotherapeutic drugs and sensitizers, suitable for in-depth mechanism exploration of targeted sensitization, drug resistance reversal and combined administration; ④ It possesses multi-dimensional malignant phenotypes including proliferation, EMT invasion, tumor stemness and abnormal activation of HER2 pathway with wide mechanism research coverage; ⑤ Its data are widely accepted by gastrointestinal oncology SCI journals. As a core benchmark model for basic and translational research of esophagogastric junction adenocarcinoma, it is applicable for methodology construction of various projects, research proposals and high-score papers.

Model Advantages

The Mouse OE19 Model is the exclusive SCI gold-standard model for targeted therapy, drug resistance mechanism research and drug screening of HER2-positive esophagogastric junction adenocarcinoma. Core advantages: With stable HER2 amplification phenotype, it serves as an irreplaceable benchmark model for targeted research in esophageal adenocarcinoma, different from ordinary gastrointestinal tumor models without HER2 amplification. It features extremely stable in vivo tumorigenesis, homogeneous proliferation and minimal batch variation with high signal-to-noise ratio and strong data reproducibility, suitable for in-depth mechanism exploration of high-score papers and innovative design of NSFC projects. It perfectly simulates the real pathological characteristics of clinical esophagogastric junction adenocarcinoma including malignant proliferation, invasion and metastasis, targeted drug resistance and recurrent progression. It can accurately carry out multi-dimensional mechanism research including HER2 pathway regulation, targeted sensitization, drug resistance reversal, EMT inhibition and stemness intervention. It can form a multi-tumor comparison system with other gastrointestinal, respiratory and urinary tumor models, enriching project levels and innovation with excellent recognition in SCI manuscripts and fund applications.

Research Applications

The Mouse OE19 Model is mainly applied to preclinical mechanism and translational research on malignant progression, targeted drug resistance, EMT invasion and metastasis, and tumor stemness of HER2-positive esophagogastric junction adenocarcinoma. Core application scenarios: analysis of HER2 amplification-mediated oncogenic pathway activation and targeted resistance mechanisms in esophagogastric junction adenocarcinoma; research on uncontrolled proliferation, EMT invasion and metastasis, and abnormal stemness activation mechanisms of esophageal adenocarcinoma; in-vivo efficacy and drug resistance reversal evaluation of HER2-targeted inhibitors, chemotherapy sensitizers, natural small-molecule compounds and nano-drug delivery systems against esophageal adenocarcinoma; exploration of synergistic efficacy and detoxification mechanisms of targeted combined chemotherapy; differentiated mechanism research on targeted therapy of gastrointestinal adenocarcinoma.
