Model Introduction

Pan02 is a murine pancreatic ductal adenocarcinoma cell line derived from C57BL/6 mouse pancreatic tissue induced by chemical carcinogen. This cell line retains key pathological features of human pancreatic cancer, including dense desmoplastic stroma, strong invasiveness and low immunogenicity. The subcutaneous xenograft model established by Pan02 cells is a widely used syngeneic preclinical in vivo model for pancreatic cancer research, extensively applied in anti-tumor drug screening, immunotherapy evaluation, tumor stroma and immune microenvironment research. Different from human tumor xenograft models, syngeneic Pan02 model is constructed on immunocompetent C57BL/6 mice, which can preserve intact murine immune system. The model has stable tumor formation, controllable growth rate and convenient real-time tumor volume measurement, suitable for large-scale in vivo pharmacodynamic screening of anti-pancreatic cancer agents.

Experimental Principle

Pan02 cells originate from C57BL/6 mouse, so the syngeneic recipient mice have no immune rejection against Pan02 tumor cells. After harvesting Pan02 cells in logarithmic growth phase and preparing cell suspension, the suspension is subcutaneously inoculated into the dorsal flank of C57BL/6 mice. Tumor cells continuously proliferate in subcutaneous tissue and eventually form solid tumor masses. Regular recording of tumor volume, body weight and survival status of mice enables assessment of in vivo tumor growth kinetics and quantitative analysis of anti-tumor efficacy of test compounds or biological agents.

Model Identification & Detection Indicators

  1. Tumor volume measurement: From the day of visible tumor formation, tumor length diameter (L) and short diameter (W) are measured every 2–3 days with vernier caliper. Tumor volume formula: TV (mm³) = (L × W²)/2.
  2. Body weight monitoring: Mouse body weight is weighed synchronously when measuring tumor volume, used to evaluate systemic toxicity induced by tumor burden or drug administration.
  3. Model success criteria: The tumor formation rate of Pan02 subcutaneous syngeneic model is expected ≥90%. Tumor volume reaches 100–150 mm³ at day 12–18 post inoculation, which is regarded as successfully modeled and available for formal grouping and intervention.
  4. Histopathological verification: At the end of experiment, tumor tissues are dissected and fixed in 4% paraformaldehyde. Hematoxylin-eosin (H&E) staining is performed to confirm pancreatic ductal adenocarcinoma pathological features; IHC staining can detect biomarkers including CK19, Ki-67, α-SMA to verify tumor origin and stroma formation activity.

Model Advantages

  1. Pan02 syngeneic model is widely cited in pancreatic cancer preclinical research, abundant literature resources and high experimental repeatability.
  2. Tumor grows steadily, forms obvious desmoplastic stroma, low spontaneous regression rate.
  3. Host C57BL/6 mice retain complete immune system, uniquely suitable for immunotherapy, immune checkpoint inhibitor and tumor immune microenvironment research.
  4. Subcutaneous inoculation procedure is mature, easy to implement, convenient for dynamic tumor size monitoring and group efficacy comparison.

Application Scenarios

  1. In vivo anti-tumor activity screening of small molecule compounds, chemotherapeutic agents, targeted drugs and immune checkpoint inhibitors against pancreatic cancer.
  2. Preclinical evaluation of tumor stroma targeted therapy, combination therapy and nanoparticle delivery system for pancreatic cancer.
  3. Mechanism study of pancreatic cancer immune suppression, tumor-stroma interaction and tumor associated fibroblast activation.
  4. Preliminary in vivo pharmacodynamic assessment before orthotopic pancreatic cancer model study.
