Model Introduction

PC-3 cell line was isolated from bone metastatic lesion of human poorly differentiated prostate adenocarcinoma. This cell line is androgen receptor (AR) negative and PSA negative, belonging to androgen-independent castration-resistant prostate cancer (CRPC), with strong tumorigenic capacity and intrinsic osteolytic bone metastasis potential. Immunodeficient mouse subcutaneous xenograft model is widely applied in preclinical anti-tumor drug screening, tumor metastasis mechanism exploration, targeted therapy and radiotherapeutic efficacy assessment.

Model Identification & Endpoint Criteria

Model Success Standard

Palpable tumor nodules can be detected at inoculation site within 14–21 days after injection. Model is considered successfully established when tumor volume reaches 100–150 mm³.

Pathological Validation

Harvest tumor tissues for H&E staining. Immunohistochemistry can detect cytokeratin and vimentin; AR and PSA show negative expression, consistent with inherent biological characteristics of PC-3 cells.

Experimental Endpoint

Terminate experiment and perform animal euthanasia when tumor volume reaches 1500 mm³, body weight loss exceeds 20%, tumor ulceration/necrosis occurs or mice exhibit poor mental activity.

Model Advantages

AR-negative and PSA-negative, androgen independent, which can well mimic advanced human CRPC, suitable for research on hormone-resistant prostate cancer.

High tumorigenicity, stable subcutaneous tumor growth, short modeling cycle and high tumor formation rate.

Possess inherent bone metastasis tropism; this model can be modified into tibial injection model for osteolytic bone metastasis research.

Widely cited in SCI publications, experimental data possess good comparability and facilitate academic paper submission.

Application Scope

Preclinical in vivo efficacy evaluation of small molecule inhibitors, chemotherapeutic agents, gene therapy, radiotherapy and combined regimens for castration-resistant prostate cancer; Mechanism research of tumor proliferation, invasion, angiogenesis and bone metastasis; Investigation of drug resistance mechanism in advanced prostate cancer.
