Modeling Principle

The Mouse RT112 Model is a dedicated gold-standard xenograft model for preclinical research on low-grade, luminal-type non-muscle invasive bladder cancer (NMIBC). Derived from clinical grade G2 human primary bladder transitional cell carcinoma, RT112 cells carry the characteristic FGFR3-TACC3 fusion gene and represent a classic FGFR3-activated bladder cancer cell line. It presents epithelial morphology, moderate differentiation, mild in-vivo proliferation, low invasiveness and stable malignant phenotypes, which faithfully recapitulate the disease characteristics and drug response patterns of clinical high-incidence low-grade recurrent FGFR3-driven bladder cancer. Logarithmic-phase RT112 human bladder cancer cells are subcutaneously inoculated into immunodeficient BALB/c-nu nude mice to establish stable, homogeneous and highly reproducible subcutaneous solid tumors without xenogeneic immune rejection. Different from highly invasive basal-type bladder cancer models, this model accurately simulates the indolent growth, high recurrence and FGFR pathway dependence of clinical low-grade bladder cancer, and is widely applied in FGFR signaling mechanism exploration, in-vivo efficacy evaluation and drug resistance research of FGFR inhibitors, chemotherapeutic agents, small-molecule natural products and nano-formulations. Due to T-cell immunodeficiency in nude mice, this model is not suitable for immune checkpoint mechanism research, but exclusively for intrinsic tumor biological research and preclinical drug screening. With a mature system and high SCI data recognition, it is fully applicable to NSFC project applications, postgraduate proposals, dissertations and standardized SCI methodology writing.

Model Validation Criteria

Quantitative Criteria for Tumor Formation and Growth

1. Positive modeling criteria: Subcutaneous tumor nodules are visible to the naked eye within 8–14 days after RT112 cell subcutaneous inoculation. The tumor formation rate is ≥90% within 2 weeks after inoculation, and local tumor volume >50 mm³ is defined as successful modeling. Tumors in the vehicle control group grow mildly and stably without spontaneous regression or explosive proliferation, which is completely consistent with the indolent growth phenotype of clinical low-grade bladder cancer. 2. Model reproducibility criteria: The intra-group coefficient of variation (CV) of tumor volume in the vehicle control group is <30%, which proves standardized modeling operation, small individual differences and excellent model data reproducibility, meeting the quality control requirements of SCI experimental data. 3. Pharmacodynamic response criteria: After intervention with FGFR-targeted positive drugs, the tumor growth inhibition rate (TGI) is ≥40%, and tumor volume and tumor weight are significantly reduced compared with the vehicle control group (P<0.05), verifying that the model has stable and sensitive pharmacodynamic response to FGFR pathway targeted intervention and chemotherapeutic drugs.

Histopathological and Molecular Gold-Standard Indexes

1. HE staining gold standard: Tumor tissues show regularly arranged epithelioid bladder cancer cells with moderate nuclear atypia, few mitotic figures and high cell differentiation, without obvious large-area necrosis, which are completely consistent with the typical pathological morphological characteristics of human low-grade G2 bladder transitional cell carcinoma. 2. Immunohistochemical phenotypic criteria: Tumor tissues present high expression of FGFR3 and GATA3, low Ki-67 proliferation index and low MMP2 expression, perfectly recapitulating the core molecular phenotypes and low-invasive, slow-proliferation biological characteristics of clinical **luminal-type, low-grade, FGFR3-mutation positive** bladder cancer. 3. Pharmacodynamic response criteria: Compared with the vehicle control group, the treatment group shows significantly down-regulated FGFR3, GATA3 and Ki-67, significantly up-regulated Cleaved-Caspase-3 and decreased MMP2 expression, indicating that drugs can effectively block the FGFR pathway, inhibit the proliferation of low-grade bladder cancer, activate tumor cell apoptosis and weaken invasive potential. 4. Evidence closed-loop criteria: In vivo histopathological and molecular protein detection results are highly consistent with the in vitro RT112 cell experimental mechanism trend, forming a complete and credible academic evidence closed loop.

Academic Evidence for Model Validity

The Mouse RT112 Model is the only exclusive gold-standard preclinical model for low-grade non-muscle invasive bladder cancer with FGFR3 fusion mutation positivity. Core academic validity evidence: ① The cell line carries typical FGFR3-TACC3 fusion gene, a high-incidence driver mutation in clinical bladder cancer, which accurately corresponds to clinically targeted therapy-sensitive populations; ② It presents classic low-grade bladder cancer phenotypes of luminal type, moderate differentiation and indolent growth, filling the research gap of highly invasive bladder cancer cell models; ③ The nude mouse xenograft system has stable tumorigenesis, high reproducibility and mild growth rhythm, suitable for long-term pharmacodynamic observation and recurrence mechanism research; ④ It is highly sensitive to FGFR-targeted drugs with clear pharmacodynamic gradients, serving as a core model for new FGFR inhibitor drug screening; ⑤ Experimental data are widely recognized by global urological oncology SCI journals. It is a key in vivo model for bladder cancer typing research, targeted drug resistance mechanism research and precise therapy transformation, suitable for various project applications, research proposals and SCI manuscript methodology writing.

Model Advantages

The Mouse RT112 Model is the most exclusive and stable preclinical gold-standard in vivo model for FGFR3-mutant low-grade luminal bladder cancer. Core advantages: It precisely carries the clinically high-incidence FGFR3-TACC3 fusion driver gene, serving as the preferred model for bladder cancer precise targeted therapy and FGFR inhibitor drug screening. As G2-grade moderately differentiated transitional cell carcinoma, it perfectly simulates the clinical characteristics of non-muscle invasive, indolent-growing and high-recurrence low-grade bladder cancer, making up for the research shortcomings of high-grade highly invasive bladder tumor models. It features mild and stable in-vivo tumorigenesis with minimal individual differences and no sudden rapid proliferation, suitable for long-acting drug evaluation and recurrence mechanism research. With extremely high phenotypic specificity, it can accurately distinguish the molecular characteristics of luminal bladder cancer, applicable for tumor typing, biomarker screening and in-depth exploration of targeted drug resistance mechanisms. With a mature system and high global SCI recognition, it forms a complete tumor research matrix with the group's MB49 immune orthotopic model and HT-1080 sarcoma model, suitable for project innovation design and high-score paper mechanism exploration.

Research Applications

The Mouse RT112 Model is mainly used for precise mechanism and targeted translational research of FGFR3-driven low-grade bladder cancer. Core application scenarios include: mechanism analysis of indolent proliferation and high recurrence of non-muscle invasive low-grade bladder cancer; research on the regulatory mechanism of downstream signaling pathways mediated by FGFR3-TACC3 fusion gene; in-vivo targeted efficacy evaluation and drug resistance mechanism exploration of single and combined FGFR inhibitor regimens; verification of the intervention effects of chemotherapy, natural small molecules and nano-drug delivery systems on luminal bladder cancer; screening of bladder cancer typing-specific biomarkers and development of precise treatment strategies. It fully supports the methodology construction of NSFC projects, postgraduate proposals, dissertations and high-score SCI papers in urological oncology, serving as an irreplaceable exclusive SCI gold-standard model for bladder cancer FGFR-targeted research.
